Fluorescence microscopy is based on the phenomenon that certain substances emit energy that can be detected as visible light when irradiated with light of a certain wavelength. The absorption and subsequent reradiation of light by organic and inorganic specimens is primarily the result of fluorescence or phosphorescence. Conventional microscopes use visible light (400-700 nanometers) to illuminate a sample and create a magnified image. Fluorescence microscopy uses a much more intense light source to excite fluorescent species in the sample of interest. This fluorescent species, in turn, emits a longer wavelength, lower energy light that produces a magnified image instead of the original light source.
