Plasmid DNA is limited by some biological barriers, such as endosomal attack, renal clearance, and degradation by serum endonucleases. In addition, GMP (Good Manufacturing Practice) grade plasmid DNA is essential for direct gene transfer to humans. Thus, to date, approximately 70% of gene therapy clinical trials involve viral vector systems.
