The isothermal amplification method provides detection of nucleic acid target sequences in a streamlined exponential manner and is not limited by the constraints of thermal cycling. These methods can be quite different, but they all share some features in common. For example, since DNA strands are not thermally denatured, all isothermal methods rely on an alternative approach that allows the initiation of primer binding and amplification reactions, i.e. polymerases with strand displacement activity. When the reaction begins, the polymerase should separate the strands that are still annealed to the sequence of interest. Isothermal amplification chemistry has been applied in diagnostics with great success and is used in many commercial molecular diagnostic platforms serving large test centers and point-of-care markets.
