Polymerase chain reaction (PCR) is a technique that makes many copies of specific DNA regions in vitro and relies on Taq polymerase, a thermostable DNA polymerase, and requires DNA primers designed specifically for the DNA region of interest. PCR enables the generation of thousands to millions of copies of specific DNA sections from very small amounts of DNA and is a common tool routinely used in DNA replication, medical diagnostics, and forensic analysis of DNA. PCR technology is based on a natural process that cells use to replicate new strands of DNA. To set up a PCR, you need 5 key ‘ingredients’: a DNA template to be copied, primers, DNA nucleotide bases, Taq polymerase enzyme, and a buffer. PCR involves a heating and cooling process called thermal cycling, which is performed by a machine in three main steps: denaturation, annealing and expansion. Repeat these 3 steps 20 to 40 times, doubling the number of DNA copies each time. After the PCR is completed, the amount and size of the generated DNA fragment can be checked by electrophoresis. PCR is used in many areas of biology and medicine, including molecular biology research, medical diagnostics, and some fields of ecology.
